HOT Solids - Material Testing

Predicting material-related responses in vivo remains a major problem in biocompatibility testing

Implant and Biomaterial Testing

Implants and biomaterials have become an integral part of regenerative medicine. Therefore, understanding how the body interacts with the material is crucial. One aspect of this is the interaction with blood, which is usually analyzed according to ISO 10993-4. However, the complex cellular interaction of the immune system with the material is often inadequately depicted. Current tests tend to focus on single types of immune cells or limited mixed populations, not allowing natural = complex cell-to-cell interactions and the investigation of a complete, in vivo-like reaction profile of the materials. Therefore, predicting material-related responses in vivo remains a major problem in biocompatibility testing.

HOT Solids

Our advanced human whole blood test system “HOT Solids” developed for solid materials bridges this gap. It features a comprehensive, physiologically relevant system that includes all blood components. Unlike static models that suffer from cellular sedimentation, our circulating or rotational test system prevents this issue, ensuring optimal contact between immune cells and the implant material while do not lead to cellular stress and immune cell activation during rotation (see Poster DGBM conference).

With incubation times up to 48 hours this provides a reproducible and highly reliable method to sensitively detect immunomodulatory effects and complex reaction profiles of implant materials after contact with human whole blood. Access to such insights during implant development will help to identify positive immunomodulatory effects (e.g. on wound healing) but also reveal undesired material properties at an early stage, resulting in significant time and cost savings (see Poster DGI/EAO conference).

Circulating Whole Blood Cultures

A new type of pump system has been developed for the examination of blood perfused medical devices (e.g. holofibre membranes, vascular implants, etc.). This minimises the mechanical stress for immune cells, but also platelets and erythrocytes.

This means that physiological conditions can be better maintained. Consequently false positive and false negative results are significantly reduced.

Summarized benefits of HOT Solids

-

- Reproducible and Reliable:

Barely any intra-donor variability effects with reliable detection of donor-dependent reactions (inter-donor variability).
	 - Prolonged Incubation Time:

Reliable insights in complex cell-to-cell interactions with an incubation period of up to 48 hours.
	 - Comprehensive Analysis:

Broad spectrum of cytokines provides detailed insight into both positive (e.g. wound healing) and potential adverse reactions. Haemocompatibility tests are also available on request.
	 - Sophisticated Test System:

Extensively tested and validated incubation methods reduce false positives and false negatives.
	 - Time and Cost Efficiency:

Early identification of problematic material properties leads to considerable time and cost savings during material development.
